Multiplexed Sequencing of Expression Cassette Expressing Human OTC in Yeast. OTC Variant Calling

Multiplexed sequencing of yeast yOTC expression cassette. yOTC encodes the human OTC protein, using a codon-optimized derivative of the human OTC cDNA sequence. Derivatives of yOTC were introduced into haploid yeast strains so that each encoded a version of human OTC with one amino acid changed. Multiplexed sequencing using the MinION platform was used to determine the altered codon, and associated amino acid change, in each yeast transformant.
