1.预处理规则:采用独创的“绿茶-白醋浸泡液”对板蓝根和茵陈进行预处理,配比规则为:100g绿茶+50ml白醋/1000ml沸水,静置30分钟后过滤,利用有机酸与多酚类物质的协同作用促进活性成分溶出。作用机理:白醋(乙酸)溶解细胞壁,绿茶多酚类物质稳定活性成分,避免氧化损失。 2.双相萃取算法:板蓝根提取:先水提(10倍水浸泡20min→煎煮2.5h→滤渣再水提2次),后醇提(60%乙醇回流3次,8倍量/2h),合并浓缩至1.10-1.20浸膏。茵陈提取:80%乙醇回流3次(10倍量/2h),浓缩至相同浸膏密度。 算法逻辑:基于极性差异分段提取,最大化捕获水溶性与脂溶性活性成分。 3.大孔树脂纯化流程:水洗7BV(柱体积)去除杂质→50%乙醇洗脱目标成分→浓缩至标准浸膏。算法目标:通过吸附-解吸机制去除无效成分,提高制剂纯度。
1. Pretreatment Protocol: An original "green tea-white vinegar soaking solution" was used to pretreat Radix Isatidis and Herba Artemisiae Scopariae. The preparation ratio is 100 g of green tea + 50 ml of white vinegar per 1000 ml of boiling water. After standing for 30 minutes, the mixture was filtered. The synergistic effect of organic acids and polyphenols was utilized to promote the dissolution of active ingredients. The mechanism of action is as follows: acetic acid from white vinegar dissolves cell walls, while green tea polyphenols stabilize the active ingredients to avoid oxidative loss. 2. Two-phase Extraction Algorithm: For Radix Isatidis extraction: Aqueous extraction was performed first: soak in 10-fold volume of water for 20 minutes, decoct for 2.5 hours, filter the residue, and perform aqueous extraction twice more. Then alcoholic extraction was carried out: reflux with 60% ethanol for 3 times, using 8-fold volume of solvent per 2 hours. The extracts were combined and concentrated to a thick extract with a density of 1.10–1.20. For Herba Artemisiae Scopariae extraction: Reflux with 80% ethanol for 3 times (10-fold volume of solvent per 2 hours), then concentrate to the same thick extract density. Algorithm logic: Stagewise extraction based on polarity differences is adopted to maximize the capture of both water-soluble and fat-soluble active ingredients. 3. Macroporous Resin Purification Process: Wash with 7 column volumes (BV) of water to remove impurities, then elute the target components with 50% ethanol, and finally concentrate to the standard thick extract. Algorithm objective: Remove inactive components via the adsorption-desorption mechanism to improve the purity of the preparation.
