肿瘤治疗过程中,如何温和、安全、靶向递送治疗剂至今仍是挑战。本研究利用合成生物学理念,创新性对外泌体的供体细胞进行基因再设计,发展了一种基于外泌体的光控RNA药物递送系统。该系统利用外泌体的生物起源以及RNA 适配体MS2-MCP蛋白相互作用、光诱导CIBN- CRY2蛋白之间的相互作用,实现了细胞内源 microRNA-21治疗剂(miR-21海绵)的高效装载和对白血病细胞的靶向递送。该研究为细胞表达的内源RNA药物提供了一种新的递送途径,并将助力更多RNA药物应用于肿瘤个性化治疗。

Currently, the development of mild, safe and targeted delivery systems for therapeutic agents remains a critical challenge in cancer therapy. In this study, we leverage synthetic biology principles to innovatively genetically reprogram exosome donor cells, and develop an exosome-based light-controlled RNA drug delivery system. This system utilizes the biological origin of exosomes, the interaction between the RNA aptamer MS2 and MCP protein, as well as the light-induced interaction between CIBN and CRY2 proteins, to achieve efficient loading of the cellular endogenous microRNA-21 therapeutic agent (miR-21 sponge) and targeted delivery to leukemia cells. This study provides a novel delivery avenue for cell-expressed endogenous RNA therapeutics, and will facilitate the application of more RNA drugs in personalized cancer therapy.
