Transcriptomic profile of hypodermal adventitia fibroblasts from Csf1f/f and Tek-crexCsf1f/f mice

In order to explore and compare the transcriptomic profile of hypodermal adventitia fibroblasts, fibroblasts were isolated via flow cytometry mouse hypodermis and were subjected to RNA-sequencing analysis. Back skins from age-matched Csf1f/f and Tek-crexCsf1f/f mice were harvested and hypodermal adventitial was isolated by mechanical force and enzymatic treatments. Cells were liberated after enzymatic digestion of the tissues. Fibroblasts (CD45- CD31- Sca-1+ CD34+) were isolated by fluorescence-activated cell sorting (FACS) and their transcriptome assessed by RNA-sequencing.
为探索并比较皮下外膜成纤维细胞的转录组谱,研究人员通过流式细胞术从小鼠皮下组织中分离成纤维细胞,并开展RNA测序(RNA-sequencing)分析。本研究收集了年龄匹配的Csf1f/f与Tek-crexCsf1f/f小鼠的背部皮肤,通过机械分离与酶消化法获取皮下外膜组织;经酶消化组织释放细胞后,通过荧光激活细胞分选术(fluorescence-activated cell sorting, FACS)分离得到表型为CD45⁻ CD31⁻ Sca-1⁺ CD34⁺的成纤维细胞,最终通过RNA测序对其转录组进行检测。
