Whole-transcriptomic analysis of circRNA-miRNA-mRNA ceRNA network in 4-month APP/PS1 mouse hippocampus

Alzheimer's disease (AD) is a progressive neurodegenerative disorder, and the molecular mechanisms of AD remain unclear. Accumulating evidence indicates the involvement of non-coding RNAs in AD pathogenesis. Here, we applied RNA-seq to explore the expression changes of circRNA, miRNA, and mRNA simultaneously, and investigate competing endogenous RNA (ceRNA) networks in hippocampus of 4-month APP/PS1 and wild-type mice. Three male APP/PS1 (APPswe and PSEN1dE9) transgenic mice and three wild type littermates were used for RNA-seq.
阿尔茨海默病(Alzheimer's disease, AD)是一种进行性神经退行性疾病,其分子发病机制尚未明确。越来越多的研究证据表明,非编码RNA参与了阿尔茨海默病的病理进程。本研究采用RNA测序(RNA-seq)技术,同时探究环状RNA(circRNA)、微小RNA(miRNA)以及信使RNA(mRNA)的表达变化,并对4月龄APP/PS1小鼠与野生型小鼠海马体中的内源竞争RNA(ceRNA)调控网络进行分析。本研究共使用3只雄性APP/PS1(携带APPswe与PSEN1dE9突变)转基因小鼠,以及3只同窝野生型小鼠开展RNA测序实验。
