Based on CD8b expression we have defined two MP-Ly49+ CD8 cells subsets. To characterize the activation of MP-Ly49+ CD8 cells during the course of an antiviral response, we performed a single-cell transcriptomic analysis. We sorted both CD8aa and CD8ab-Ly49+ cells from the spleen of naive or VV-infected mice at 8 days post-infection. We also sorted Ly49-CD44+ and naive cells from naive mice and B8R-specific cells from VV-infected mice, as controls.
