该品系在小鼠 Gfap 启动子作用下,在神经星形胶质细胞特异表达 Cre 酶。中靶小鼠可 作为神经星形胶质细胞特异性诱导 LoxP 重组的 Cre 工具鼠。例如删除 FLOX 区域,即将该 工具鼠与得到的条件性敲除模型小鼠配繁,可在神经星形胶质细胞特异性删除两个 LoxP 之 间的基因片段。 通过 Cas9 技术定点整合至 H11 位点,而 H11 位点作为 KI 的插入位点具有显著的优势: 1.转入的基因定点整合至染色体,故传代稳定,不会出现因传代导致表达丢失的现象;2.能 避免基因组上毗邻序列的干扰效应;3. 外源基因插入到这个位置不会破坏任何内源性基因, 并且小鼠生长发育正常、不影响小鼠表型及功能。 H11位点位于小鼠11#染色体,如果将此基因敲入小鼠与其他小鼠品系配繁,请尽量避免两个靶基因位于同一染色体上,否则将无法得到双基因阳性同时敲入纯合的小鼠后代。

This mouse strain specifically expresses Cre recombinase in neural astrocytes under the control of the mouse Gfap promoter. Targeted mice can serve as Cre driver mice for specifically inducing LoxP recombination in neural astrocytes. For example, to delete a FLOX-flanked genomic region, this Cre driver mouse can be crossed with conditional knockout mouse models, enabling the specific deletion of the gene fragment flanked by two LoxP sites in neural astrocytes. This transgene is site-specifically integrated into the H11 locus via Cas9-mediated genome editing. The H11 locus, as a knock-in (KI) insertion site, has remarkable advantages: 1. The integrated exogenous gene is stably integrated into the chromosome, ensuring stable germline transmission without expression loss caused by successive breeding generations; 2. It avoids interference effects from adjacent genomic sequences; 3. Insertion of exogenous genes at this locus does not disrupt any endogenous genes, and the mice develop normally without affecting their phenotypes and physiological functions. The H11 locus is located on mouse chromosome 11. When crossing this knock-in mouse with other mouse strains, please avoid placing both target genes on the same chromosome; otherwise, no mouse offspring that are homozygous for both targeted knock-in alleles will be generated.
