In order to investigate the effect of parental age at fertilization on the mutation rate, we have generated a tissue bank from consecutive litters between two crosses (129S/B6 and B6/129S) and several breeding mouse pairs. We intend to use this resource to investigate whether there is an age effect in the maternal and paternal lines, and if so, establish the nature of the effect. We are also interested in examining the differences in mutation rates between strains, and investigating the variance among contemporaneous gametes. Initially, we aim to sequence the parents and 5 pups from the oldest and youngest litter, and from two different crosses. This will require deep WGS (>25X after duplicate removal). Once the data is generated, we will also mine the data to study germline and somatic mosiacism in the parents and offspring. This study will complement work already being undertaken on the mutation rate in humans and radiation exposed mice.
