<i>C. auris, </i>an emerging fungal pathogen, preferentially colonizes human skin and causes outbreaks of systemic infections. However, the factors that regulate the pathogenesis of <i>C. auris</i> are still unclear. Using unbiased scRNA-seq profiling of murine skin infected with <i>C. auris</i>, we identified the host immune factors that are potentially involved in the regulation of <i>C. auris</i> in the skin. Furthermore, our <i>in vivo</i> scRNA-seq reveals that <i>C. auris</i> infection upregulates IL-1Ra in the myeloid cells and limits neutrophil antifungal activity. In addition, our results suggest that <i>C. auris</i> mannan layer can evade IL-1R signaling mediated skin defense. Collectively, our findings identified the immune and non-immune cells involved in the regulation of <i>C. auris</i> in the skin which could open the door to gain insights into the pathogenesis of this emerging fungal pathogen.
