RNF12 E3 ligase activity-dependent transcriptional profile in mouse embryonic stem cells

RNF12 is a developmental E3 ligase mutated in Tonne-Kalscheuer Syndrome patients. To identify the gene expression programme controlled by RNF12 activity in mouse embryonic stem cells we employ RNA-Sequencing. Overall design: RNF12 knockout embryonic stem cells transfected with: 1) empty vector, 2) RNF12 wild type, and 3) RNF12 W576Y catalytic mutant. Total RNA was extracted, libraries generated and subjected to RNA-sequencing in an Illumina NextSeq 500 instrument. FastQ files were generated using the BaseSpace inbuilt BCL to FastQ workflow.
