We aimed to determine the relative contributions of MyD88- and TRIF-dependent signaling in Toll-like receptor-induced trained immunity in macrophages. Overall design: Bone marrow-derived macrophages were obtained from wild type (WT), MyD88 KO (MKO), and TRIF KO (TKO) mice (3 biological replicates each) and treated with the TLR4 agonist monophosphoryl lipid A (1ug/mL MPLA; _M) or left unstimulated as negative controls (_US) for 24h before washoff. Macrophages were then rested for 3 days before RNA was harvested and processed for RNAseq.
