Laser Capture Microdissection (LCM) RNA sequencing of transgenic rice lines overexpressing OsCGA1

CYTOKININ-RESPONSIVE GATA FACTOR 1 (CGA1) is a key regulator of chloroplast biogenesis, to evaluate the impact of specific expression of OsCGA1 in rice bundle sheath cells using FtGLDp promoter, we seperated bundle sheath strands and mesophyll using laser capture microdissection and performed RNA sequencing. Consistent with enhance chloroplast development, differential gene expression analysis suggests that ectopic expression of OsCGA1 is sufficient to mediate a transcriptional cascade leading to a global increase in photosynthetic machinery in rice bundle sheath cells.
