The lupins exhibit a remarkable variation of their chromosome number (2n = 32 to 52) and their genome size (2C = 1 to ~ 2.6 Gb). In order to understand the evolutionary dynamics of the lupine genome, a comparative NGS-based evaluation of the repetitive DNA compartment has been conducted on ten species representing the main clades known in the genus Lupinus and one outgroup (Ulex minor). For this purpose, a sequence dataset of 120 Mb (1,200,000 PE reads) per accession, randomly extracted from resources generated by low-depth genomic Illumina HiSeq sequencing (representing 5 to 10% of each genome), was analyzed with the program RepeatExplorer (Novak et al., 2013), which allows detection, quantitative estimation and annotation of repetitive sequences in NGS data.
