Integration of the pSLT plasmid into the Salmonella chromosome

In a screen for mutations affecting NAD metabolism, a rare mutant was isolated in which the pSLT virulence plasmid had inserted into the sppA gene of the Salmonella genome. Replication initiated from the pSLT plasmid resulted in a gradient in read coverage when the genome of the mutant was sequenced. Insertion of pSLT replication origins at other chromosomal sites and in other chromosomes could be a useful tool for studying replication and large chromosomal rearrangements.
