Ppm1d T/+ mice exhibit altered BM composition. The aim is to determine the molecular mechanisms possibly responsible for this alterations in sorted long-term hematopoietic stem cells isolated from bone marrow. Long-term HSCs (LT-HSCs sorted from 12-week-old WT and Ppm1DT/+ mice. The Ppm1dT/+ HSC transcriptome shows enrichment of MYC, mTOR compared to WT.
Ppm1d T/+ 小鼠表现出骨髓(Bone Marrow, BM)组成异常。本研究旨在阐明分离自骨髓的分选纯化长期造血干细胞中,介导该骨髓组成改变的潜在分子机制。实验从12周龄野生型(Wild Type, WT)与Ppm1d T/+小鼠中分离得到长期造血干细胞(Long-term Hematopoietic Stem Cells, LT-HSCs)。相较于野生型小鼠,Ppm1d T/+小鼠造血干细胞的转录组中,MYC与哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin, mTOR)呈现富集现象。
