RNA-seq of A375 cells treated with I-BET726 alone and sequential combination of I-BET726 and Oxaliplatin

Sequential therapy is a promising approach to enhance antitumor efficacy; however, rational design of sequential drug treatments remains challenging. Here, we applied an AI framework, AlphaTherapy, to identify a specific sequential drug combination of I-BET726 and Oxaliplatin that exhibits antitumor effects in melanoma (A375 cells). To investigate the molecular mechanisms underlying the antitumor activity of this combination, gene expression was profiled by RNA sequencing (RNA-seq) after treatment. Overall design: RNA-seq was performed on A375 cells under the following conditions: untreated control (DMSO), 2 µM I-BET726 for 72 h, withdrawal treatment (2 µM I-BET726 for 72 h followed by 72 h without drug), and sequential treatment (2 µM I-BET726 for 72 h followed by 10 µM Oxaliplatin for 72 h).
