Individuals adopt distinct behavioral coping strategies when exposed to stress. These coping strategies are essential for survival and underlie the differential susceptibility to stress-related disorders. The roles of corticotropin-releasing factor (CRF) and the medial prefrontal cortex (mPFC) in stress-related behavioral responses have been extensively studied, yet the function of CRF-expressing neurons in the medial prefrontal cortex remains unclear. This study employed morphological, electrophysiological, and calcium imaging approaches and found that CRF neurons in the medial prefrontal cortex are a subtype of GABAergic inhibitory neurons that directly participate in tail suspension stress responses. Apoptotic ablation or chemogenetic inhibition of CRF neurons in the dorsomedial prefrontal cortex increased immobility time during tail suspension and forced swim challenges, and induced social avoidance behaviors, whereas activation of these CRF neurons produced the opposite behavioral effects. Furthermore, enhancing the activity of CRF neurons promoted individual resilience to repeated social defeat stress. Collectively, these results reveal the critical role of medial prefrontal cortex CRF neurons in bidirectionally regulating behavioral selection under stressful conditions.
