162 genes were identified as conferring resistance to 2-amino-3-methylimidazo[4,5-f]quinoline (IQ) and 334 genes were identified as conferring resistance to bioactivated IQ. Overall design: Two pooled diploid yeast deletion collections, one original and one expressing human CYP1A2 and NAT2, were exposed to either 400 μM IQ, 800 μM IQ, 2% methanol (+0.1% acetic acid), or nothing, for either 5 or 10 generations. Barcode DNA was extracted from pooled samples and differential abundance calculated for each strain in the collections to determine which deletion strains were sensitive to IQ and which were resistant.
