Transcriptomic profile of M0 and M1 phenotype macrophages.

Bone marrow derived macrophages from 8-week-old C57BL/6 mice was stimulated ex vivo with or without LPS and IFN-? for 24 hours. Each group has two replicates. Overall design: Bone marrow cells were collected, washed, and resuspended in complete DMEM media containing 1% penicillin–streptomycin and 10% FBS to a concentration of 1 × 106 cells/ml. Bone marrow cells were differentiated into macrophages for 7 days in the presence of 20ng/ml recombinant murine MCSF in complete medium, with complete media changes every 48 hours. On day 6, macrophages were cultured for 24 hours at 37°C and 5% CO2 in complete media containing 20ng/ml recombinant MCSF alone (M0) or in the media containing 20 ng/mL IFN-? and 100 ng/mL LPS (M1).
