Mitochondrial genome displays considerable heterogeneity within an individual or even a single tissue. Evidence suggests that de novo mtDNA mutations may have functional significance during stem cell dysfunction and aging. A significant hurdle to new insights into the roles of mitochondria in these processes is our lack of understanding of mitochondrial genome integrity at the single-cell level. To overcome these hurdles, we developed a novel method to sequence individual mtDNA within single cells, termed single-cell individual Mitochondrial Genome sequencing (iMiGseq). We applied iMiGseq to do mtDNA sequencing of single human and mouse oocytes.
