Simultaneous targeting linked loci in mouse embryos using base editing

Base editing covers an exceptional range of applications, both in basic and applied sciences. Through the co-injection of two base editors and two sgRNAs into mouse zygotes, we introduced C•G-to-T•A transitions into two putative cytokine-sensing transcription enhancers separated by 9kb. Whole genome sequences (WGS) was conducted to investigate possible off-target mutations on predicted off-target sites.
