This study aimed to report the first cases of Candida auris isolation at the largest hospital in northern Portugal in 2023, linked to both candidaemia and colonization. Strain identification, antifungal susceptibility profiling, and genome sequencing analysis were conducted. All strains from patients with candidaemia or colonization exhibited high MIC values for fluconazole and amphotericin B. These strains were classified as Clade I (South Asian). The isolates harbored the ERG11 (Y132F) and CDR11 (E709D) mutations, along with a newly identified FKS1 (I1465L) mutation.
