N6-Methyladenosine (m6A) is the most prevalent internal modification in mammalian mRNAs, and participates in various fundamental bioprocesses as well as cancer. Here we immunoprecipitated m6A methylated poly (A+) RNAs (MeRIP) and then sequenced and profiled mRNA m6A methylation in P493-6 cells. Overall design: To investigate the effects of MYC on mRNA m6A modification, we used the human P493-6 B cell model of Burkitt’s lymphoma. P493-6 cells carry a MYC tetracycline (Tet)-off system, which enables the generation of cells with low or high MYC expression. P493-6 cells that were treated with Tet for 72 hours or 0 hour with 1 ug/ml Tet.
