本研究通过添加不同通路抑制剂，探讨特定代谢途径对皮肤干细胞增殖和体外分化的影响。通过对比基质细胞是否参与的培养条件，证明与基质细胞共培养可以建立稳定的多层皮肤样器官结构。最后，建立了由人类胚胎干细胞衍生的多种皮肤细胞组成的类皮肤器官。研究中，通过形态学观察、HE染色和荧光染色分析细胞组成。This study investigated the effects of specific metabolic pathways on the proliferation and in vitro differentiation of skin stem cells by adding various pathway inhibitors. By comparing culture conditions with or without stromal cell involvement, it was confirmed that co-culture with stromal cells can establish stable multi-layered skin-like organoid structures. Finally, skin-like organoids composed of multiple skin cell types derived from human embryonic stem cells were established. Cellular composition was analyzed via morphological observation, hematoxylin-eosin (HE) staining, and fluorescence staining.
