Type I interferon upregulates proteins that inhibit HIV replication within infected cells. Here, we interrogate interferon-stimulated gene (ISG) restriction of HIV in primary CD4+ T cells by performing comprehensive CRISPR-knockout screens using a custom library that specifically targets ISGs expressed in CD4+ T cells (CD4-ISG library). Screens were performed in two donors (Donor 1, Donor 2) with two HIV strains (Q23.BG505, LAI) at two MOIs (MOI 0.1, MOI 1) for a total of eight CRISPR screens. We utilized the HIV-CRISPR screening approach (Ohainle et al, 2018) which requires deep sequencing of genomic DNA (gDNA) and viral RNA (vRNA) for each screen. Therefore, there are 32 sequencing files in this dataset.
