This study investigates the transcriptomic effects of PRAME knockdown in the A375SM skin cutaneous melanoma cell line. Cells were transduced with either control shRNA (shCtrl) or PRAME-targeting shRNA (shPRAME), followed by paired-end RNA sequencing. The dataset provides gene expression changes associated with PRAME silencing and offers insights into PRAME-regulated pathways in melanoma. Overall design: A total of six RNA-seq samples were generated using the A375SM melanoma cell line: three biological replicates transduced with control shRNA (shCtrl) and three replicates transduced with PRAME-targeting shRNA (shPRAME). Total RNA was extracted, polyA-enriched, and sequenced using paired-end 150 bp reads on an Illumina NovaSeq platform. Raw FASTQ files and processed gene expression matrices are provided for differential expression analysis.
