To investigate the accumulation of non coding small RNAs we performed high throughput RNA sequencing on size selcted total RNA from malaria parasite Plasmodium falciparum Overall design: Total RNA was used with the NEBNext small RNA seuqencing kit to create a sequencing library with size selction <100nt. Resulting sequencing library was mapped to organellar genomes (apicoplast and mitochondria) to determine clusters of organellar small RNAs. In a second experiment, RNA from enriched Plasmodium falciparum organelles was subjected to the same library preparation, sequencing scheme, and mapping analysis.
