Metabolomic Analysis Reveals the Mechanisms Underlying Ischemic Stroke Induced by Vitamin B12 Deficiency
Objective: This study aims to explore the impact of vitamin B12 deficiency on the pathogenesis of ischemic stroke through a comprehensive analysis combining metabolomics and proteomics methodologies. Methods: Patients experiencing their first episode of acute ischemic stroke (AIS) will be recruited and categorized into two groups based on the presence or absence of vitamin B12 deficiency, alongside a matched healthy control cohort. An extensive untargeted metabolomic investigation will be conducted using ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) to detect differentially expressed metabolites (DEMs) among the groups. The protein expression profiles will be analyzed using 4D-DIA proteomics technology to identify significantly differentially expressed proteins (DEPs). Subsequently, an integrated analysis of the identified DEMs and DEPs will be performed to unveil potential biomarkers and metabolic pathways implicated in the pathogenesis of ischemic stroke.
